Prior to the launch of Sekiro: Shadows Die Twice, From Software teased an encounter with a giant snake called the Great Serpent. The encounter with the Great Serpent doesn't really play out like a traditional Sekiro boss battle, but the beast presents its own threat to the player, forcing them to get past it in order to continue the story.

Preparing to Kill the Snake

When players first encounter the Great Serpent, their main concern will be getting past the snake. Some players may be wondering if it's actually possible to kill the snake in Sekiro instead of just getting past it, and as it turns out, yes you can kill the snake.

Anyone interested in killing the snake in Sekiro will first need to have the Mortal Blade. The Mortal Blade itself isn't important to killing the snake, but the other ability players learn alongside getting the Mortal Blade is. After getting the Mortal Blade in Sekiro, players will learn the Puppeteer Ninjutsu, which they have to use in order to get to a platform above the snake.

How to Kill the Snake

After getting the Puppeteer Ninjutsu, Sekiro: Shadows Die Twice players need to head to the Senpou Temple - Mt. Kongo Sculptor's Idol save point. From here, they should be able to find a kite, with a rat-like creature near the controls. Using the Puppeteer Ninjutsu on the rat (deathblow from behind, then press the right bumper again), players will be able to raise the kite and use it to progress to the ledge above the snake.

Continue through the cliff side using the grappling hook and eventually there will be a cave with a Sculptor's Idol near it. Use the save point if necessary and then go through the cave to finally reach the ledge above the snake. Look down at the snake and jump off the ledge to deliver the first deathblow. Afterwards, there should be an option to deliver a followup deathblow to kill the snake in Sekiro.

What You Get for Killing the Snake

After killing the snake, Sekiro players will get a couple of rewards. For one, they will earn the Fresh Serpent Viscera item. And secondly, they will also unlock of Sekiro's achievement/trophies for going through the trouble of killing the snake.
