In this area, you will be taking on Doopliss, an evil Duplighost who has turned all the inhabitants of the town into pigs. To reach this location, you will have to find the correct route from Rogueport.

How to Find Twilight Town Entrance
In order to reach Twilight Town, you will first need to complete Chapter 3 in Glitzville, where you took on a bunch of fighters in the Glitz Pit, and defeated Grubba in a final battle.
When you return to Rogueport after visiting Professor Frankly, make your way to the west side of the town, past the area where you stepped on Zess T.'s contact lens. In this location, head to the very center, where there will be a storm drain. Use your Paper Mode ability, which allows Mario to turn sideways, so that you can slide through the holes in the drain.
Once you are underneath Rogueport, bring out your Yoshi and use him to float across to the area on the left side with the green pipe. Head through the blue door at the back, where there will be a brown pipe inside. However, the pipe will reject you. Return to Professor Frankly in Rogueport to tell him about the situation.

How to Find Darkly
After chatting with Professor Frankly, he will tell you that you'll need to find Darkly, who lives in the area. Head back outside, and next to the house with the Shine Sprite sign above it, head straight to the wall and turn right, where you'll be able to slip behind the homes. You will find Darkly at the back, next to a puddle.
Darkly will write Mario's name and his partner's name on their clothes, allowing you access through the pipe to Twilight Town. Now, return to the western side of town and slip through the storm drain again, and enter through the Brown Pipe where you'll successfully be allowed through. You'll arrive in Twilight Town, where you'll need to prepare to take on Doopliss and use some Stylish Moves while you're at it.
